ERG20-(GGGS)3-MRBBS
The ERG20-(GGGS)3-MRBBS assembly is a composite part for the effective production of α-biasobolol in Saccharomyces cerevisiae. It is a combination of CDS ERG20 Part:BBa_K849001 and MRBBS via a flexible linker (GGGS)3. The ERG20 expresses farnesyl pyrophosphate synthetase, which catalyzes the biosynthesis of farnesyl pyrophosphate (FPP). The MRBBS expresses α-bisabolol synthase, which catalyzes FPP into α-biasobol. The (GGGS)3 is a flexible linker tested and selected from various linker sequences, which was proved to improve the production of α-biasobolol when fusing ERG20 and MRBBS. The ERG20-(GGGS)3-MRBBS was fused with a GAL10 promoter upstream, which triggers the production of α-biasobolol at the presence of GAL4 protein. It may be regulated by other kinds of promoters for other experimental purposes. To optimize the production of α-biasobolol, the original ERG9, which catalyzes FPP into squalene, should be knocked down. However, because ERG9 is essential for the viability of Saccharomyces cerevisiae, it should not be completely knocked out.
